Two beta-glucosidases from the legumes Dalbergia cochinchinensis and Dalbergia nigrescens were compared for their ability to hydrolyze isoflavonoid glycosides from soybean. Both D. nigrescens and D. cochinchinensis beta-glucosidases could hydrolyze conjugated soybean glycosides, but D. nigrescens beta-glucosidase hydrolyzed both conjugated and nonconjugated glycosides in crude soybean extract more rapidly. The kinetic properties Km, kcat, and kcat/Km of the Dalbergia beta-glucosidases toward conjugated isoflavonoid glycosides, determined using high-performance liquid chromatography, confirmed the higher efficiency of the D. nigrescens beta-glucosidase in hydrolyzing these substrates. The D. nigrescens beta-glucosidase could also efficiently hydrolyze isoflavone glycosides in soy flour suspensions, suggesting its application to increase free isoflavones in soy products.  相似文献

We examined the mechanism responsible for low reproductive success in leatherback turtles (Dermochelys coriacea) at Playa Grande, Costa Rica: low egg fertilization versus high rates of embryonic death. Leatherbacks at this beach had a high rate of fertility (=93.3%±2.5%, n=819). We incubated 10 eggs from every clutch encountered of 19 females during 3 months of the 1998-1999 nesting season. Fertility rate of some females decreased during the nesting season, but overall was high. Detection of fertility was difficult using standard methods because fertility rates cannot be determined accurately from nests excavated after hatching because of egg decomposition. Removal and incubation of eggs from nests provided a better estimate. Embryonic death, particularly in the beginning of incubation before embryos are visible to the unaided eye, was the cause of low hatching success in this population. Hatching success increased with increasing fertility and differed between females, with some mothers having 71-81% success and others 23-32%. Embryonic death and not low egg fertility drives poor recruitment at Playa Grande. Improved conservation of this species at Playa Grande will require a better understanding of the mechanism behind embryonic death.  相似文献

The relationship between atmospheric SO2 concentrations and rainwater pH is examined for a coastal locality in North Merseyside, England for the period 1978 to 1982. During this period rainwater pH has risen slightly but significantly while atmospheric SO2 levels have fallen. The relationship between SO2 levels, rainwater pH and wind frequency and wind run from eight sectors of the compass are analyzed. Higher SO2 concentrations coincide with southerly, southeasterly and easterly winds from urban and industrial source areas. Slightly higher frequency and run of wind occur from the south and east during the winter season when SO2 production is increased.  相似文献

Near-infrared reflectance spectroscopy (NIRS) was evaluated as a possible alternative to gas chromatography (GC) for the quantitative analysis of fatty acids in forages. Herbage samples from 11 greenhouse-grown forage species (grasses, legumes, and forbs) were collected at three stages of growth. Samples were freeze-dried, ground, and analyzed by GC and NIRS techniques. Half of the 195 samples were used to develop an NIRS calibration file for each of eight fatty acids, with the remaining half used as a validation data set. Spectral data, collected over a wavelength range of 1100-2498 nm, were regressed against GC data to develop calibration equations for lauric (C12:0), myristic (C14:0), palmitic (C16:0), stearic (C18:0), palmitoleic (C16:1), oleic (C18:1), linoleic (C18:2), and alpha-linolenic (C18:3) acids. Calibration equations had high coefficients of determination for calibration (0.93-0.99) and cross-validation (0.89-0.98), and standard errors of calibration and cross-validation were < 20% of the respective means. Simple linear regressions of NIRS results against GC data for the validation data set had r2 values ranging from 0.86 to 0.97. Regression slopes for C12:0, C14:0, C16:0, C18:0, C16:1, C18:2, and C18:3 were not significantly different (P = 0.05) from 1.0. The regression slope for C18:1 was 1.1. The ratio of standard error of prediction to standard deviation was > 3.0 for all fatty acids except C12:0 (2.6) and C14:0 (2.9). Validation statistics indicate that NIRS has high prediction ability for fatty acids in forages. Calibration equations developed using data for all plant materials accurately predicted concentrations of C16:0, C18:2, and C18:3 in individual plant species. Accuracy of prediction was less, but acceptable, for fatty acids (C12:0, C14:0, C18:0, C16:1, and C18:1) that were less prevalent.  相似文献

A plot study was conducted to quantify long-term (>12 years) impacts of a single biosolids application, and short-term impacts (<2 years) of a repeated application, on semi-arid rangeland soil chemical and microbial parameters. In 2003 and 2004, plots which had received 0, 2.5, 5, 10, 21, or 30 Mg biosolids ha−1 once in 1991 (long-term plots), or again in 2002 (short-term plots), were sampled and analyzed for soil chemical parameters, microbial biovolumes, C and N mineralization activities, Biolog EcoPlate substrate utilization potential, and plant productivity and tissue quality. Repeated applications temporarily exacerbated differences in soil chemical properties among treatments, but after 2 years, soil chemistry trends were similar between short-term and long-term plots. Soils which received a repeated application of 21 or 30 Mg biosolids ha−1 had greater bacterial biovolumes and C and N mineralization activities. In long-term plots, mineralization activities were stimulated only at the highest rate. Biosolids-amended soil communities also utilized Biolog substrates more quickly compared to communities from control plots. Plant biomass increased, whereas plant diversity and plant C/N ratio decreased with increasing application rate for both short- and long-term plots. Infrequent biosolids application had positive ecosystem effects in terms of site management objectives, with relatively low extractable metal levels in soil and greater plant biomass and tissue quality despite reduced species richness.  相似文献

The objective of the present study was to determine whether concentrations of different isoflavones (puerarin, genistein, genistin, daidzein, and daidzin) in shoots and roots of five selected soybean genotypes would respond the same or differently to red (650 nm peak transmittance) and far-red (750 nm peak transmittance) light treatments given under controlled environments. Levels of isoflavones (mg g(-1) dry weight biomass) present in seeds, control roots, and shoots and 10 day light-treated seedlings (light, dark, red, and far-red wavelengths) of soybean (Glycine max) were determined by high-performance liquid chromatography analysis in comparison with known isoflavone standards. Seeds of the five soybean genotypes studied consistently stored most of their isoflavones as glucosyl conjugates (e.g., daidzin, genistin, and puerarin). For the five soybean genotypes, isoflavone levels were lower in the seeds as compared with roots plus shoots of control, time zero (first true leaf stage) seedlings. Following 10 days of the respective light treatments, we found that (i) isoflavone levels were enhanced in dark-grown plants over light-grown plants for three of the five genotypes (a new finding) and the reverse occurred for a single genotype (a typical response of legumes) and (ii) generally, far-red end of day (EOD) light treatment enhanced total isoflavone levels in roots plus shoots over red EOD light treatment. Results from the present study show that phytochrome does appear to play a role in regulating isoflavone levels in developing soybean seedlings and that this influence by red/far-red-mediated phytochrome reactions is strongly dependent on the genotypes selected for study.  相似文献

Isocupressic acid (1) was used to synthetically prepare a mixture of (8S,13R,S)-labda-15,19-dioic acid (tetrahydroagathic acid) (5) via a two-step oxidation procedure followed by hydrogenation of the double bonds at C13 and C8. Reduction of the C8,17 double bond was stereospecific producing only the 8S isomer and confirmed by the nOe interaction between the resulting C17 and C20 methyl groups. The 13R and 13S isomers of 5 were separated and analyzed by HPLC/MS, and (13S)-tetrahydroagathic acid was isolated and identified by comparison to a standard prepared by hydrogenation of naturally occurring (13S)-dihydroagathic acid (4). (13R,S)-dihydroagathic acid was prepared by selective sodium metal-catalyzed hydrogenation of the C13,14 allylic double bond of agathic acid (3). The prepared compounds were then used as standards to confirm the presence of 4 and 5 and their respective 13R and 13S isomers in bovine serum samples. Tetrahydroagathic acid was shown to be the only metabolite detected in serum samples taken from a suspected cattle abortion case submitted for diagnosis; and, thus, 5 could be a valuable diagnostic marker for pine needle-induced abortions.  相似文献

The onset of age-related neurodegenerative diseases such as Alzheimer's or Parkinson's disease, superimposed on a declining nervous system, could exacerbate the motor and cognitive behavioral deficits that normally occur in senescence. In cases of severe deficits in memory or motor function, hospitalization and/or custodial care would be a likely outcome. This means that unless some way is found to reduce these age-related decrements in neuronal function, health-care costs will continue to rise exponentially. Thus, it is extremely important to explore methods to retard or reverse age-related neuronal deficits, as well as their subsequent behavioral manifestations, to increase healthy aging. In this regard, consumption of diets rich in antioxidants and anti-inflammatory polyphenolics, such as those found in fruits and vegetables, may lower the risk of developing age-related neurodegenerative diseases. Research suggests that the polyphenolic compounds found in berry fruits, such as blueberries and strawberries, may exert their beneficial effects either through their ability to lower oxidative stress and inflammation or directly by altering the signaling involved in neuronal communication, calcium buffering ability, neuroprotective stress shock proteins, plasticity, and stress signaling pathways. These interventions, in turn, may exert protection against age-related deficits in cognitive and motor function. The purpose of this paper is to discuss the benefits of these interventions in rodent models and to describe the putative molecular mechanisms involved in their benefits.  相似文献

UV spectral fingerprints, in combination with analysis of variance-principal components analysis (ANOVA-PCA), can differentiate between cultivars and growing conditions (or treatments) and can be used to identify sources of variance. Broccoli samples, composed of two cultivars, were grown under seven different conditions or treatments (four levels of Se-enriched irrigation waters, organic farming, and conventional farming with 100 and 80% irrigation based on crop evaporation and transpiration rate). Freeze-dried powdered samples were extracted with methanol-water (60:40, v/v) and analyzed with no prior separation. Spectral fingerprints were acquired for the UV region (220-380 nm) using a 50-fold dilution of the extract. ANOVA-PCA was used to construct subset matrices that permitted easy verification of the hypothesis that cultivar and treatment contributed to a difference in the chemical expression of the broccoli. The sums of the squares of the same matrices were used to show that cultivar, treatment, and analytical repeatability contributed 30.5, 68.3, and 1.2% of the variance, respectively.  相似文献
